Yes, you need to keep the one x to keep using kinect. Series x does not recognize the kinect adapter. I would keep my one x anyways because I heard xbox one games cannot be installed offline on the series x due to drm. Not a problem now, but it could be many years from now.
That's not what the setting does. The chat mixer was originally intended for Kinect environments, then to a lesser extent when stereo chat headsets were available. What the setting does is adjust the balance of TV and game/party chat that comes over the speakers when there is no headset connected to the controller/console.
